Abstract

enObjective: To evaluate the therapeutic potential of bumetanide in autism spectrum disorder (ASD) by analyzing its neurobiological rationale, preclinical evidence, and results from randomized controlled clinical trials. Methods: A critical review of the literature was conducted, including five randomized controlled trials, preclinical studies, and pharmacological data identified through PubMed and institutional sources. Results: Small and medium-sized studies suggest a moderate clinical improvement associated with modulation of the GABAergic system. In contrast, large phase III trials did not demonstrate a significant clinical benefit at the population level, and no global cognitive improvement was observed. Conclusion: Bumetanide does not represent a universal treatment for ASD but may constitute a targeted therapeutic approach for specific subgroups of patients, warranting more precise patient selection and further investigation.
