Abstract
Migraine is a recurring and highly disabling neurological disorder that can worsen if not treated properly. However, most of the drugs suggested against migraine attacks or as a long-term treatment don’t have an ideal profile. Some can cause medication overuse headaches while others have side effects and contraindications that often limit their use. Botulinum toxin, a natural protein complex synthesized by Clostridium botulinum, has been used for decades in various neuromuscular disorders. It’s also believed to be effective in preventing chronic migraine, according to the results of two placebo-controlled phase III clinical trials published in 2010. This thesis aims to determine whether botulinum toxin is a relevant prophylactic agent in the treatment of chronic migraine. Its efficacy and safety, analyzed in a recent meta-analysis and reassessed in long-term studies, are discussed in this work. The place of botulinum toxin in the therapeutic arsenal of migraine is also supported by the study of its mechanism of action at the neuronal level.
